Polyethylene glycol (abbreviated as PEG) is a synthetic polymeric material; its main chain repeatedly consists of two methylenes and an oxygen atom. It is soluble in water and some organic solvent.
PEG can be connected to some molecules and their surface by chemical reaction to form protective membrane compatible with lives. The membrane minimizes the exclusiveness of material against biological system, material surface absorption of proteins, cells and pathogens and excretion speed from kidney.
When connected with high molecule weight active PEG, the drug changes in the following aspects:
Improve the solubility and stability of drug; reduce or eliminate the immunogenicity; decrease the metabolism speed of drug; slow down excretion speed from renal glomerulus for a bigger size of drug; the mechanism of some carbohydrates and peptide compounds to accept and clean up drugs is passivated because of the cover of PEG over the drug; the administration interval is longer for its longer circulation time; improve drug distribution to minimize its side effects.
The potential usages include: surface modifying; liposome modifying; the connection between molecules and molecules or between molecules and their surface; biological purification; the synthesis of biomacromolecules and improve the solubility.
